بررسی خاصیت ضد باکتریایی(in vitro) برخی گیاهان دارویی بر جدایه های بیماری زای استرپتوکوکوس اینیایی

چکیده

استرپتوکوکوس اینیایی عامل بیماری استرپتوکوکوزیس، یک پاتوژن زئونوز است که باعث خسارات زیادی به صنعت آبزی پروری در سراسر جهان از جمله ایران گردیده است. در مطالعه کنونی اثرات ضدباکتریایی اسانس های آویشن باغی(Thymus vulgaris)، پونه (Mentha longiflora) و زیره سیاه (Carum carvi) بر دو جدایه بیماری زای استرپتوکوکوس اینیایی با روش رقیق سازی در محیط مایع و جذب نوری در طول موج 600 نانومتر بررسی و سپس با روش دیسک با آنتی بیوتیک های اریترومایسین و انروفلوکساسین مقایسه شدند. بر اساس نتایج حداقل غلظت مهار رشد (MIC) اسانس آویشن باغی، پونه و زیره سیاه برای این باکتری به ترتیب 8/0، 8/0 و 6/1-8/0 میکرولیتر بر میلی لیتر و همچنین حداقل غلظت باکتری کشی(MBC) اسانس ها به ترتیب 8/0، 6/1 و 6/1میکرولیتر بر میلی لیتر می باشد. بعلاوه براساس نتایج جذب نوری روند رشد جدایه های باکتری با افزایش غلظت اسانس ها کاهش یافته بطوریکه در غلظت های MIC و بالاتر تقریبا رشد آنها متوقف شده است. همچنین در روش دیسک میزان قطر هاله عدم رشد ناشی از اسانس آویشن باغی بطور معنی داری بیشتر از دیگر اسانس ها و آنتی بیوتیک‌های اریترومایسین و انروفلوکساسین بود. بطور خلاصه نتایج این آزمایش بیانگر خاصیت ضد باکتری مناسب آویشن باغی بوده و پیشنهاد می‌گردد برای کنترل و درمان بیماری استرپتوکوکوزیس در مزارع قزل‌آلا ارزیابی گردد.

The zoonotic bacteria, Streptococcus iniae is the causative agent of streptococcosis associated with serious economic losses in the word aquaculture industry. The antimicrobial activities of Thymus vulgaris, Carum carvi, and Menthalong flora essential oils were determined against two pathogenic isolates of S. iniae using broth microdilution, and were compared with erythromycin and enrofloxacin antibiotics by disk diffusion assay. Also, bacterial growth was followed by OD at 600 nm. The minimum inhibitory concentration (MIC) of the T. vulgaris, C. carvi, and M. flora essential oils were 0.8, 0.8, and 0.8-1.6 μl/ml, and the minimum bactericidal concentration (MBC) values were found as 0.8, 1.6, and 1.6 μl/ml, respectively. Moreover, the growth of isolates was reduced by increasing essential oils concentration. Also, disk diffusion assay revealed that the antimicrobial activity of T. vulgaris essential oil is significantly higher than the other plants, as well as erythromycin and enrofloxacin antibiotics. These findings support the use of T. vulgaris for the control of streptococcosis in trout farms.
