But over time, the resistant bacteria to this antibiotic were identified one after another. Antibiotics of the penicillin family have a β-lactam ring in their molecular structure. By competitively binding to transpeptidase, penicillin and other β-lactam antibiotics inhibit its activity. Inhibition of transpeptidase activity prevents cell wall synthesis and causes bacterial death. Resistant bacteria, have β-lactamase enzyme which has the ability to break the C-N bond in the β-lactam ring. In this study, we reconstructed a network of known β-lactamase genes in E.coli. The results showed that ampC gene plays a central role in induction of resistance in E.coli. Molecular function analysis of other β-lactamase genes showed that β-lactamase activity is only a secondary function for them and their main function is to participate in other unrelated cellular processes. Gene ontology analysis showed that the process of resistance to β-lactam antibiotics was positively correlated with electron transport chain. Also, when exposed to β-lactam antibiotics, the cell membrane is the most active cellular component in E.coli and antibiotic resistance is dependent on the β-lactamase function of cell membrane proteins.}, keywords = {Antibiotics,Beta-Lactam,gene network,Bacteria}, title_fa = {بازسازی شبکه ژنی مقاومت به پنی‌سیلین در E.coli با استفاده از اطلاعات موجود در پایگاه‌های داده}, abstract_fa = {کشف پنی‌سیلین، انقلابی را در صنعت پزشکی ایجاد کرد و جان هزاران نفر را نجات داد. اما با گذشت زمان باکتری‌های مقاوم به این آنتی‌بیوتیک یکی پس از دیگری شناسایی شدند. آنتـی‌بیوتیک‌های خانواده پنی‌سیلین در ساختـار مولکولی خود دارای یک حلقه‌ی بتا–لاکتامهستنـد. پنی‌سیلین و دیگر آنتی‌بیوتیک‌های دارای حلقه بتا-لاکتام با اتصال به آنزیم ترانس‌پپتیداز به صورت رقابتی فعالیت آن را مهار می‌کنند. مهار آنزیم ترانس‌پپتیداز از سنتز دیواره باکتریایی جلوگیری کرده و سبب مرگ باکتری می‌گردد. باکتری‌های مقاوم دارای آنزیم بتا-لاکتاماز هستند که توانایی شکستن پیوند C-N را در حلقه بتا-لاکتام دارند. در این مطالعه بازسازی شبکه ژن‌های شناخته شده بتا-لاکتاماز در باکتری E.coli صورت گرفت. نتایج نشان داد ژن ampC نقش محوری در القای مقاومت در E.coli دارد. بررسی عملکرد مولکولی سایر ژن‌های شبکه بتا-لاکتامازی نشان داد ویژگی بتا-لاکتامازی برای آنها، صرفاً یک قابلیت ثانویه بوده و وظیفه اصلی آنها مشارکت در سایر فرایند‌های غیر مرتبط سلولی می‌باشد. بررسی ژن آنتولوژی نشان داد فرایند مقاومت به آنتی‌بیوتیک‌های بتا-لاکتام همبستگی مثبت با زنجیره انتقال الکترون دارد. همچنین بررسی جایگاه سلولی ژن‌ها نشان داد غشای سلولی فعال‌ترین اندامک سلولی در زمان مواجهه با آنتی‌بیوتیک‌های بتا-لاکتام در باکتری E.coliمی‌باشد و مقاومت به آنتی‌بیوتیک وابسته به عملکرد بتالاکتامازی پروتئین‌های فعال در غشای سلولی است.}, keywords_fa = {آنتی‌بیوتیک,بتا-لاکتام,شبکه ژنی,باکتری}, url = {https://nfvm.uoz.ac.ir/article_105952.html}, eprint = {https://nfvm.uoz.ac.ir/article_105952_7bb3e38144055e0fae74b7e9db1f5d2f.pdf} }
