In intrinsic resistance, the bacteria show resistance due to their specific properties to one or all of the antibiotics. But acquired resistance is caused by molecular changes in susceptible bacteria and, eventually, it causes antibiotic-resistant bacteria to emerge. That could be due to chromosomal mutations, transposons, or transmissible plasmids. Numerous genes control the different mechanisms of bacterial resistance to antibiotics. The tetA and tetB genes are the major activating genes for tetracycline efflux mechanism, which decrease the concentration of tetracycline in bacterium. This study was performed to reconstruct the network of tetA and tetB genes using data and information in molecular databases. The reconstructed network clearly confirms that tetA and tetB genes have a direct role in tetracycline efflux. In association with other proteins, tetB, in addition to tetracycline efflux, plays a key role in the detoxification and antiport of a wide range of toxins, such as streptocycline and phenolic acids. Gene Anthology analysis showed that most of the genes involved in the tetracycline resistance process are membrane proteins and their molecular function is transporter.}, keywords = {Antibiotics,Bacteria,gene network,Tetracycline}, title_fa = {بررسی شبکه ژنی مقاومت به آنتی بیوتیک تتراسایکلین تحت کنترل ژن‌های tetA و tetB با استفاده از اطلاعات موجود در پایگاه‌های‌داده}, abstract_fa = {فرایند مقاومت به آنتی بیوتیک به دو دسته مقاومت ذاتی و مقاومت اکتسابی تقسیم می‌شوند. در مقاومت ذاتی باکتری به دلیل ویژگی‌های خاص خود به یک یا تمام آنتی بیوتیک‌ها مقاومت نشان می‌دهد. اما مقاومت اکتسابی در اثر تغییرات مولکولی در باکتری‌های حساس به یک آنتی بیوتیک ایجاد شده و نهایتا سبب بوجود آمدن باکتری‌های مقاوم به آن آنتی بیوتیک می‌شود که دلیل آن می‌تواند جهش‌های کروموزومی، ترانسپوزون‌ها و یا پلاسمیدهای قابل انتقال باشند. ژن‌های متعددی مکانیسم‌های متنوع مقاومت باکتریایی را نسبت به آنتی بیوتیک‌ها کنترل می‌کنند. ژن‌های tetA و tetB اصلی‌ترین ژن‌های فعال کننده مکانیسم پمپ یونی افلاکس تتراسایکلین بوده و سبب کاهش غلظت تتراسایلکین در داخل باکتری می-شوند. در این مطالعه شبکه ژن‌های tetA و tetB با استفاده از داده‌ها و اطلاعات موجود در پایگاه‌های داده‌های مولکولی بازسازی شد. شبکه بازسازی شده به روشنی تایید می‌کند که ژن‌های tetA و tetB در افلاکس تتراسایکلین به خارج از سلول نقش مستقیم دارند. ژن tetB با همکاری سایر پروتئین‌ها، علاوه بر افلاکس تتراسایکلین، نقش کلیدی در سمیت زدایی و آنتی‌پورت طیف گسترده‌ای از سموم مثل استرپتوترسین و اسیدهای فنولیک دارد. همچنین آنالیزهای ژن آنتولوژی نشان داد. اغلب ژن‌های درگیر در فرایند افلاکس تتراسایکلین پروتئین غشایی هستند و عملکرد مولکولی آن‌ها ترانسپورتر می‌باشد.}, keywords_fa = {آنتی بیوتیک,باکتری,تتراسایکلین,شبکه ژنی}, url = {https://nfvm.uoz.ac.ir/article_97352.html}, eprint = {https://nfvm.uoz.ac.ir/article_97352_7a3b04b8c008d6e8478afab8d963bdce.pdf} }
