اثرات ضد میکروبی عصارههای هندوانه ابوجهل و کلپوره بر برخی باکتریهای بیماریزای
در طول تاریخ بشری، بسیاری از بیماریهای عفونی بهطور سنتی با داروهای گیاهی درمان شدهاند، بهطوریکه امروزه در بسیاری از کشورهای در حال توسعه، داروهای گیاهی نقش اصلی را در درمان اولیه ایفا میکنند. هدف از این تحقیق، بررسی اثرات ضد میکروبی عصارههای اتانولی، متانولی و اتیل استات هندوانه ابوجهل (میوه) و کلپوره (برگ) بر برخی باکتریهای بیماریزا بوده است. گیاهان مورد بررسی از شهرستان زابل جمعآوری شده، سپس آسیاب شده و در حلالهای اتانول، متانول، اتیل استات قرار داده و در نهایت عصارهگیری با دستگاه روتاری انجام شد. باکتری با جمعیت استاندارد تهیه شده و حداقل غلظت مهارکنندگی و حداقل غلظت کشندگی با روش میکرودایلوشن بر روی باکتریهای ویبریو کلرا، استافیلوکوکوس اورئوس، باسیلوس سرئوس، شیگلا دیسنتری و لیستریا مونوسیتوژنز تعیین شد. کمترین غلظت مهارکنندگی عصاره اتانولی (5/12 پیپیام)، متانولی (25 پیپیام) و اتیل استات (25 پیپیام) هندوانه ابوجهل به ترتیب بر باکتریهای لیستریا مونوسیتوژنز، ویبریو کلرا و باسیلوس سرئوس مؤثر بودهاند. عصاره اتانولی کلپوره با کمترین غلظت 50 پیپیام تنها توانسته باکتری ویبریو کلرا را مهار کند. کمترین غلظت مهارکنندگی عصاره متانولی کلپوره (25 پیپیام) در برابر باکتری ویبریو کلرا مشاهده و باکتری باسیلوس سرئوس در تمام غلظتها رشد کرده است. کمترین غلظت مهارکنندگی عصاره اتیل استات با 5/12 پیپیام بر باکتری ویبریو کلرا مشاهده شده است. در کل عصاره اتانولی هندوانه ابوجهل بر باکتری لیستریا مونوسیتوژنز و عصاره اتیل استات کلپوره بر باکتری ویبریو کلرا مؤثر بودهاند لذا پیشنهاد میگردد بسته به هدف استفاده از عصاره گیاهی از نوع خاص گیاه به همراه مؤثرترین حلال مربوطه استفاده شود.

تأثیر محلول روئی عاری از سلول Lactobacillus reuteri بر میزان رشد گونه توکسینزای قارچ Fusarium oxysporum در آزمایشگاه
لاکتوباسیلوس روتری یک پروبیوتیک است که در حضور گلیسرول، روترین را که یک ماده ضد میکروب با طیف گسترده است تولید میکند. مطالعه حاضر با هدف ارزیابی اثربخشی محلول روئی عاری از سلول لاکتوباسیلوس روتری بر رشد قارچ فوزاریوم اوکسیسپوروم انجام گرفت. بدین منظور، محلول روئی عاری از سلول باکتری، از سانتریفیوژ (rpm 8000 به مدت 15 دقیقه) کشت اورنایت باکتری لاکتوباسیلوس روتری در محیط با و بدون گلیسرول، تهیه شد. اثر مهاری آن با استفاده از دو روش: میکروتیترپلیت (مقادیر سریالی از 100 تا 25/6 میکرولیتر از دو نوع محلول روئی عاری از سلول؛ 10 میکرولیتر اسپور قارچ (spores/ml 106×2)؛ محیط PDB) و روش کشت در لوله (مقادیر 60 و 80 مایکرولیتر از دو نوع محلول روئی عاری از سلول؛10 میکرولیتر اسپور قارچ (spores/ml 106×2)؛ محیط SDB) بررسی شد. حداقل غلظت بازدارندگی و حداقل غلظت کشنده محلول روئی عاری از سلول حاصل از محیط کشت حاوی دو درصد گلیسرول، 60 میکرولیتر تعیین گردید. محلول روئی عاری از سلول حاصل از محیط کشت فاقد گلیسرول، اثر مهاری بر رشد قارچ فوزاریوم اکسیسپوروم نشان نداد. با توجه به نتایج مطالعه حاضر محلول رویی عاری از سلول بهدست آمده از محیط کشت لاکتوباسیلوس روتری حاوی دو درصدگلیسرول احتمالاً به دلیل حضور روترین، قادر به مهار رشد گونه توکسینزای فوزاریوم اکسیسپوروم بوده و میتواند بهعنوان یک عامل کنترلکننده زیستی بالقوه علیه گونههای توکسینزای فوزاریوم، در صنایع خوراک دام و مواد غذایی، مورد استفاده قرار گیرد.

بررسی ژنوتیپ و مشخصههای ملکولی سویههای ویروس نکروز عفونی پانکراس شناسایی شده در برخی مزارع قزلآلای کشور
بیماری نکروز عفونی پانکراس ناشی از ویروسIPNV است که سویههای مختلف آن دارای حدت متفاوتی مرتبط با برخی موتیف های کپسید VP2 میباشند. در مطالعه حاضر نمونههای مشکوک به بیماریIPN از مزارع قزلآلا در سالهای 2018 تا 2019 بررسی و نمونههای مثبت تعیین ژنوتیپ شده و از نظر حدت ملکولی مورد ارزیابی قرار گرفتند. ویروس در نمونههای مزارع استان های کرمانشاه و همدان با استفاده از واکنش RT-PCR ردیابی گردید. براساس آنالیز توالی آنتیژنVP2 ، سویههای شناسایی شده متعلق به ژنوتیپ 5 و سروتیپ Sp بوده و با همدیگر و با سایر سویههای ایرانی و همچنین سویههای گزارش شده در ترکیه و برخی کشورهای اروپایی بیش از 99 درصد شباهت دارند. همچنین سویههای ایرانی شناسایی شده در این مطالعه دارای اسیدهای آمینه پرولین و ترئونین در موقعیت 217 و 221 بودند که معمولاً در سویههای با حدت کم دیده میشوند. به طور خلاصه میتوان عنوان نمود که سویههای شناسایی شده در برخی مزارع قزلآلای کشور (استانهای همدان و کرمانشاه) متعلق به ژنوتیپ 5 و سروتیپ Sp (A2) با منشأ اروپایی میباشندکه علیرغم داشتن موتیف P217T221 دارای حدت متوسط میباشند و احتمالاً از طریق واردات تخم چشم زده به کشور وارد شده اند.

تاثیر سیر تازه در جیره روی جمعیت اشریشیاکلی روده و برخی از فاکتورهای بیوشیمیایی سرم در جوجههای گوشتی
هدف از این تحقیق ارزیابی اثرات سیر تازه در جیره روی جمعیت اشریشیاکلی و برخی از فاکتورهای بیوشیمیایی سرم خون در جوجههای گوشتی بود. در مجموع 105 جوجه در شهرستان زابل برای این تحقیق استفاده شدند. در روز هفتم 96 جوجه با وضعیت بهتر انتخاب و بهطور تصادفی به 3 گروه مساوی با 2 تکرار تقسیم شدند. جیرهی گروههای 1 (کنترل)، 2 و 3 (درمان) بهترتیب با مقادیر 0، 1 و 2 درصد سیر تازه مخلوط گردید. در سن 35 روزگی از هر تکرار 8 جوجه بهطور تصادفی انتخاب و وزن شدند و از آنها نمونه خون تهیه گردید و سپس کشتار شدند. پس از کالبدگشایی غده لنفاوی بورس فابریسیوس از بدن خارج و وزن گردید. برای شمارش تعداد پرگنههای اشریشیاکلی در روده از محتویات ایلئوم استفاده شد. نتایج بررسی نشان داد جوجههایی که سیرتازه در جیره دریافت کرده بودند افزایش معنیداری در وزن بورس فابریسیوس را نسبت به گروه کنترل نشان دادند (05/0P<). تفاوت معنیداری برای جمعیت اشریشیاکلی، میزان کلسترول، تریگلیسرید و پروتئین تام بین گروه کنترل و گروههای درمان مشاهده نگردید (05/0P>).

بررسی تاثیر نانو کمپلکس قارچ و مس بر روی باکتری های ایجاد کننده ی عفونت بیمارستانی
عفونت بیمارستانی یکی از عمدهترین و اساسیترین مشکلات درمانی در تمام بیمارستانها تلقی میگردد و به طور میانگین 5 تا 10 درصد بیماران بستری را درگیر مینماید. از عوامل تشدید کنندهی عفونت بیمارستانی میتوان به استفادهی مکرر از ابزارها و همچنین تماسهای پیدرپی با کادر درمانی اشاره نمود. در این مطالعه اثرات ضد باکتریایی نانوذرات مس با استفاده از عصارهی آبی قارچ گانودرما لوسیدوم بر روی سوشهای استاندارد باکتریهای استافیلوکوکوساورئوس، اسینتوباکتربومانی و استرپتوکوکوس پایوژنز و سودوموناس آئروژینوزا که عامل ایجاد کنندهی عفونتهای بیمارستانی میباشند مورد بررسی قرار داده شد. جهت اندازهگیری ابعاد و شکل نانو ذرات مس از میکروسکوپ الکترونی استفاده گردید. همچنین جهت بررسی ترکیبات آلی احتمالی که در سنتز نانو ذرات امکان دخالت را داشتند آنالیز طیف سنجی مادون قرمز انجام شد. جهت مشاهدهی میزان اثربخشی این نانوکمپلکس تست MTT بر روی دو ردهی سلولی vero و ردهی سلولی فیبروبلاستL 929 صورت پذیرفت. نتایج میکروسکوپ الکترونی روبشی این پژوهش نشان داد که نمونه عصاره دارای مورفولوژی کاملاً یکنواخت میباشد. نانو ذرات مس کروی و در محدوده 30-20 نانومتر میباشند. همچنین در بررسی تست mtt آزمونANOVA نشان داد که مقدارSig برابر صفر است که نشان دهندهی تفاوت معنیدار میان غلظتهایمختلف و نمونههای شاهد مثبت و منفی وجود دارد. طبق نتایج بهدست آمدهی این مطالعه نانو کمپلکس قارچ و مس دارای اثر ضد باکتریایی قوی علیه باکتریهای مولد عفونتهای بیمارستانی بوده است.

بررسی تأثیر ضد میکروبی عصارههای الکلی آلوئهورا، بابونه آلمانی و نعناع فلفلی بهعنوان نگهدارنده طبیعی شیر پاستوریزه
اخیراً تحقیقات در خصوص استفاده از ترکیبات گیاهی بهعنوان نگهدارندههای طبیعی در غذاها مورد توجه قرار گرفته است. هدف از این پژوهش، ارزیابی فعالیت ضد میکروبی عصارههای متانولی و اتانولی سه گیاه دارویی آلوئهورا، بابونه آلمانی و نعناع فلفلی کشت شده در پژوهشکده کشاورزی دانشگاه زابل بر میکروبهای عامل فساد و بیماریزای شیر پاستوریزه شامل سودوموناسآئروژینوزا، باسیلوسسرئوس، میکروکوکوسلوتئوس، لیستریامنوسایتوژنز و اشرشیاکلی و در مرحله بعد بررسی اثر عصارههای اتانولی آلوئهورا و بابونه آلمانی بر ماندگاری شیر پاستوریزه بود. نتایج نشان داد که گیاه آلوئهورا بیشترین اثر ضد میکروبی و گیاهان بابونه آلمانی و نعناع فلفلی به ترتیب بعد از آن قرار داشتند (05/0≥P). به طور کلی عصاره اتانولی گیاهان مورد آزمایش دارای اثر ضد میکروبی قویتری نسبت به عصاره متانولی بود (05/0≥P). غلظتهای ۱۵/۰، ۳/۰ و ۶/۰ (درصد حجمی/حجمی) از عصارههای اتانولی آلوئهورا و بابونه آلمانی، بهعنوان نگهدارنده طبیعی در شیر پاستوریزه مورد ارزیابی قرارگرفت. نتایج حاکی از آن بود که تیمار شیر پاستوریزه با ۳/۰ و ۱۵/۰ درصد بابونه آلمانی و همچنین آلوئهورا با غلظت ۳/۰ درصد ضمن داشتن خصوصیات حسی قابل قبول، بهطور معنیداری جمعیت میکروبی پایینتر و زمان ماندگاری طولانیتری در مقایسه با نمونه شاهد داشت. بنابراین استفاده از عصاره گیاهان مذکور بهعنوان نگهدارنده در شیر پاستوریزه با خصوصیات مفید یک غذای عملگرا پیشنهاد میشود.

تعیین میزان شیوع استرپتوکوکوس اکوئی تحتگونه زواپیدمیکوس در جنینهای سقط شده مادیان به روش مولکولی
سقط جنین در دامهای اهلی همواره بهعنوان یکی از معضلات صنعت دامپروری در تمام نقاط جهان مطرح، که عوامل مسبب آن متعدد و متنوع است. استرپتوکوکوس اکوئی تحتگونه زواپیدمیکوس یک کوکسی گرم مثبت و کاتالاز منفی است که یکی از علل مهم سقط و از دست دادن کره در هنگام آبستنی، در مادیان به حساب میآید. هدف از این مطالعه بررسی حضور ژنوم این باکتری در تعدادی از جنینهای سقطی مادیان در استانهای غربی کشور بود. برای این منظور 125 نمونه محتویات آسپیره شده معدههای سقط شده با روش PCR برای شناسایی ژن sodA مورد آزمایش شد. نتایج این مطالعه نشان داد که 23 (4/18 درصد) مورد از 125 نمونه جنین سقط شده با استرپتوکوکوس اکوئی تحتگونه زواپیدمیکوس آلوده بودند. یافتههای مطالعه حاضر نشان دهنده حضور بالایی از عفونت استرپتوکوکوزیس بوده و نشان میدهد که یکی از علل مهم سقط جنین در مادیان بوده و برنامههای کنترلی برای کاهش ضررهای اقتصادی این باکتری در ایران ضروری است. در این مطالعه برای اولین بار یک باکتری در محتویات معده جنینهای سقط شده مادیان بهعنوان یکی از علل اصلی سقط جنین در ایران تشخیص داده شد.

مروری بر کنترل و درمان جرب قرمز طیور (Dermanyssus gallinae)
جرب قرمز طیور، مهمترین انگل خارجی خونخوار در گلههای تخمگذار و مادر در بسیاری از کشورها است. آلودگی با این جرب اجباری، بسیار رایج بوده و بر اساس گزارشات اپیدمیولوژیک، 83 درصد از مزارع اروپایی به آن آلوده هستند. همچنین، درمانیسوس گالینه بهعنوان رایجترین و مهمترین آفت طیور در ایران توصیف شده است. آلودگی با جرب قرمز طیور، میتواند منجر به کاهش تولید تخممرغ، استرس، سرکوب سیستم ایمنی، پرکنی، کانیبالیسم، آنمی و مرگ شود. به علاوه، اثبات شده است که این جرب، میتواند برخی از عوامل بیماریزا از جمله سالمونلا را منتقل کند. از این گذشته، آلودگی انسان به جرب درمانیسوس گالینه به صورت فزایندهای از کشورهای مختلف از جمله ایران، گزارش شده است. اگرچه روشهای گوناگونی برای کنترل این جرب در سالنهای طیور گزارش شده، اما رویکرد اصلی، متکی بر استفاده از ترکیبات ضد کنه سنتتیک میباشد. اهداف این مطالعهی مروری، بررسی جنبههای گوناگون آلودگی با جرب درمانیسوس گالینه، توصیف ترکیبات ضد کنه مختلف و ارزیابی اثرات هر کدام از ترکیبات ضد کنه بر روی این جرب میباشند.

بررسی فنوتیپی مقاومت آنتیبیوتیکی در جدایههای کورینه باکتریوم سودوتوبرکلوزیس جدا شده از گوسفندان مبتلا به لنفادنیت پنیری شهرستان بینالود مشهد
لنفادنیت پنیری یک بیماری عفونی پوستی بسیار شایع در بین گوسفندان و بزها در سراسر جهان است که عامل این بیماری باکتری کورینه باکتریوم پسودوتوبرکلوزیس است. ایجاد این بیماری در بین دامها همواره ضررهای اقتصادی چشمگیری به همراه دارد. در این مطالعه تعداد 40 نمونه از آبسههای موجود در موارد دام زنده و لاشههای کشتارگاه دام در شهرستان مشهد و بینالود مورد بررسی قرار گرفت. جهت نمونهگیری از عقدههای لنفاوی دارای تورم و چرک، به وسیله سرنگ نمونه از داخل غدد متورم به مقدار حدود 1 سیسی جمعآوری شد و سرنگ حاوی نمونه در کنار یخ به آزمایشگاه انتقال داده شد. نمونههای جمعآوری شده به محیط کشت BHI براث انتقال داده شدند و پس از تأیید توسط تستهایی از قبیل رنگآمیزی گرم ، تست کاتالاز و تشخیص از روی پرگنه باکتری، دیسکگذاری جهت تعیین میزان مقاومت و حساسیت جدایهها به روش کربی بائر انجام شد. بررسی تعیین میزان حساسیت و مقاومت در جدایههای مورد مطالعه نشان داد که بیشترین میزان مقاومت جدایهها نسبت به آنتیبیوتیک داکسی سایکلین (5/72 درصد) بود و کمترین میزان مقاومت نسبت به آنتیبیوتیکهای سولفامتوکسازول تریمتوپریم (5/17 درصد) بود. روش دیسک دیفیوژن آگار میتواند به عنوان یک روش غربالگری اولیه و ابتدایی جهت تعیین میزان مقاومت و حساسیت استفاده گردد لذا برای بررسی دقیق میزان مقاومت جدایهها میتوان از روش ژنوتیپی استفاده کرد.

مروری بر بیماری و شیوع ویروس اسهال ویروسی گاو (Bovine Viral Diarrhea Virus) در ایران
بیماری اسهال ویروسی گاوان (BVD) در اکثر نقاط دنیا بومی شده و خسارت عمده این بیماری اعم از درگیری دستگاههای مختلف بدن، کاهش تولید شیر، مشکلات تولید مثلی مانند سقط جنین، کاهش وزن، کاهش رشد و تضعیف سیستم ایمنی میباشد. شیوع بالا و همچنین انتقال سریع بین دامها از خصوصیات این بیماری است که اهمیت کنترل آن را چند برابر کرده است. شرایط گاوداریها، وضعیت جغرافیایی مناطق، وجود فاکتورهای مستعدکنندهی دیگر از جمله وجود دیگر بیماریهای ویروسی و باکتریایی در محل نگهداری دامها و سیاست مدیریتی حاکم بر میزان زایش و تولید شیر همگی از عوامل مؤثر بر شیوع بیماری اسهال ویروسی گاوان است. در ایران مطالعات زیادی شیوع این بیماری را در مناطق مختلف گزارش کردهاند. بهطوری که بیشترین شیوع این بیماری به ترتیب در شهرهای مشهد، البرز و کرمان گزارش شده است. میانگین درصد شیوع بیماری اسهال ویروسی گاوان در ایران بنابر مطالعات انجام گرفته تا اواخر سال 1399، برابر با 37/47 درصد است که شیوع و گستردگی فراوان این بیماری را در سراسر کشور نشان میدهد.
