ارزیابی واکسن تهیه شده از باکتری کشته شده Vibrio fluvialis در مرغ تخم گذار نژاد هایلاین W36
باکتری Vibriofluvialisموجب بیماری در ماهیان پرورشی و ضررهای اقتصادی قابل توجه میشود. این باکتری پاتوژن انسانی و چندین نوع از ماهیان پرورشی دریایی میباشد. استفاده از تکنولوژی ایمونوگلوبولین زرده تخممرغ (IgY) تا کنون برای کنترل عفونتهای میکروبی فراوانی مورد استفاده قرار گرفته است. هدف از مطالعه حاضر بررسی و ارزیابی واکسن تهیه شده از باکتری V. fluvialisپس از تزریق به مرغ تخمگذار میباشد. باکتری V. fluvialis پس از کشت در محیط مایع، با فرمالئین 0.05% مجاور شد. سپس سلول باکتری به همراه اجوانت فروند به عنوان واکسن به مرغهای تخمگذار نژاد هایلاین در 7 نوبت تزریق شد، افزایش وزن مرغها تا بلوغ جسمی و تولید تخممرغ مورد ارزیابی قرار گرفت، سپس آزمایش الایزا غیر مستقیم برای شناسایی IgG ضد باکتری V. fluvialisطراحی و اجرا شد. تزریق واکسن تهیه شده در افزایش وزن مرغها در طول آزمایش و تعداد و وزن تخممرغها معنی دار نبود و آزمایش الایزا نشان داد تیتر آنتیبادی در سرم مرغهای هایپر ایمن شده پس از تزریق چهارمین بوستر به 1:12800 رسیده است. با توجه به نتایج بدست آمده و نتایج مطالعات سایر محققین بنظر میرسد، آنتیبادی زرده تخممرغ (IgY) که همان IgG منتقل شده به زرده است، توانایی مهار بیماری زایی این باکتری را در حیوان آزمایشگاهی و مزرعه دارد.

بررسی آلودگی به انگل کریپتوسپوریدیوم در بوقلمون های بومی شهرستان بابل
کریپتوسپوریدیوم از جمله بیماریهای مهم در دام، طیور و انسان است که هم از نظر بهداشتی و هم از لحاظ اقتصادی دارای اهمیت فراوانی میباشد. این بیماری توسط انگلهای جنس کریپتوسپوریدیوم ایجاد میشود که یک تکیاخته انگلی از شاخه اپی کمپلکسا است. این انگل منجر به اختلال در دستگاه گوارش و بروز اسهال یا درگیری دستگاه تنفس میگردد.مطالعه حاضر به منظور بررسی میزان آلودگی به انگل کریپتوسپوریدیوم در بوقلمونهای بومی شهرستان بابل در سال 1395 انجام گرفت.در این بررسی، تعداد 200 نمونه مدفوعی از بوقلمونهای بومی تهیه و با روش ذیل نلسون مورد آزمایش قرار گرفت. بر اساس نتایج بدست آمده از 200 نمونه مورد بررسی، در 15% نمونهها آلودگی به انگل کریپتوسپوریدیوم مشاهده شد. جنس نر و ماده از لحاظ میزان شیوع آلودگی اختلاف معنیداری نداشتند. میزان آلودگی در فصل پاییز بیشتر از دیگر فصلها بود.نتایج حاصله از این بررسی نشان داد که آلودگی به کریپتوسپوریدیوم در بوقلمونهای بومی شهرستان بابل نسبتا زیاد میباشد، لذا لازم است مطالعات بیشتری در این زمینه انجام گرفته و راهکارهای مناسبی در رابطه با پیشگیری و کنترل این بیماری ارائه گردد.

بررسی تغییرات تیتر آنتی بادی بر علیه بیماری نیوکاسل و آنفلوانزا در زرده تخم مرغ در دوره انکوباسیون و تاثیر آن بر تکثیر داخل تخم مرغی ویروس ها
آنتیبادیهای مادری از طریق زرده تخممرغ جذب شده و وارد گردش خون جوجه میشوند. ایمنی پاسیو کسب شده از آنتیبادی مادری در جوجه میتواند اثرات عوامل عفونی را کاهش داده یا از بروز بیماریهای بالینی جلوگیری کند. بیماری نیوکاسل و آنفلوانزا از بیماریهای بسیار مهم و واگیر در پرندگان است که خسارات اقتصادی زیادی به صنعت طیور وارد میکنند. تست ممانعت از هماگلوتیناسیون یکی از تستهای رایج و عمومی برای تشخیص تیتر آنتیبادی بر علیه ویروس نیوکاسل و آنفلوانزا در پرندگان است. در بررسی حاضر در سه مرحله مقادیر تیتر آنتیبادی بر علیه بیماری نیوکاسل و آنفلوانزا در سرم مرغ مادر، زرده تخممرغ و جوجه یک روزه به روش تست ممانعت از هماگلوتیناسیون اندازهگیری گردید. در این بررسی در سه دوره متوالی تیتر آنتیبادی نیوکاسل و آنفلوانزا در سرم مرغ مادر اندازهگیری و با مقادیر آن در زرده تخممرغ حاصل از همان گله مقایسه گردید. مقادیر تیتر این آنتیبادیها در زرده تخممرغ در دوره انکوباسیون در روزهای اول، سوم، ششم، نهم، چهاردهم و هیجدهم اندازهگیری شد. همچنین این مقادیر با تیتر آنتیبادی در سرم جوجه یک روزه همان گروه تخممرغها مقایسه گردید. در قسمتی از بررسی با تزریق ویروسهای نیوکاسل و آنفلوانزا در حفره آلانتوئیک تخممرغ جنین دار مقادیر تست EID50 در تخممرغهای دارای آنتیبادی نیوکاسل و آنفلوانزا با مقادیر آن در تخممرغهای فاقد آنتیبادیهای مذکور((SPF مقایسه گردید. بررسی آماری نتایج نشان داد که مقادیر آنتیبادی نیوکاسل و آنفلوانزا در سرم خون مادر تفاوت معنیداری با مقادیر این آنتیبادی در زرده تخممرغ این گله ندارد (P˃0.05). نتایج اندازهگیری مقادیر آنتیبادی نیوکاسل و آنفلوانزا در دوره انکوباسیون نشان داد که مقدار این آنتیبادی در مدت انکوباسیون در روزهای مختلف و همچنین بین مقادیر آنها در زرده و سرم جوجه یک روزه تفاوت معنیداری وجود ندارد (P˃0.05). بنابراین میتوان با اندازهگیری آنتیبادی در زرده تخممرغ، تیتر آنتیبادی را در سرم گله مادر و سرم جوجه حاصل از این تخممرغها تخمین زد. نتایج تست EID50 نشان داد که EID50 ویروس نیوکاسل، حاصل از تزریق آن به تخممرغ SPF نسبت به تزریق آن به تخممرغ دارای آنتیبادی بالاتر بوده (P<0.05) ولی در مورد ویروس آنفلوانزا تفاوت معنیداری بین مقادیر EID50 در تخممرغ SPF و تخممرغ دارای آنتیبادی وجود ندارد (P˃0.05). لذا در مورد نیوکاسل جهت انجام تست مذکور بهتر است از تخممرغ SPF استفاده گردد.

بررسی ﻣﻘﺎوﻣﺖ آﻧﺘﯽ ﺑﯿﻮﺗﯿﮑﯽ جدایه های کلبسیلا پنومونیه به دست آمده از ﻧﻤﻮﻧﻪ ﻫﺎی ﺑﺎﻟﯿﻨی بیمارستان خاتم الانبیاء (ص) زاهدان طی سال های 92-1391
کلبسیلاپنومونیه ازجمله باکتریهای عامل عفونت ادراری و یکی از پاتوژنهای فرصت طلب و عامل عفونت بیمارستانی محسوب میگردد. شیوع مقاومت دارویی کلبسیلا پنومونیه روزبهروز رو به افزایش است و بنابراین انجام تستهای مقاومت دارویی، قبل از تجویز آنتیبیوتیک، ضروری به نظر میرسد. هدف این مطالعه تعیین میزان مقاومت به برخی از آنتیبیوتیکها در ایزولههای کلبسیلا پنومونیه جدا شده از نمونههای بالینی بیماران بیمارستان خاتم الانبیاء (ص) زاهدان طی سالهای 1391 تا 1392 با استفاده از روش انتشار دیسک بود. در ﻣﻄﺎﻟﻌﻪ ﺗﻮﺻﯿﻔﯽ- ﺗﺤﻠﯿﻠﯽ ﺣﺎﺿﺮ̨ تعداد 83 جدایه ی کلبسیلاپنومونیه از ﻧﻤﻮﻧﻪ ﻫﺎی ﻣﺨﺘﻠﻒ ﺑﺎﻟﯿﻨﯽ در بیمارستان خاتم جداسازی و ﺑﺎ اﺳﺘﻔﺎده از آزﻣﻮنهای ﺑﯿﻮﺷﯿﻤﯿﺎﯾﯽ ﺗﻌﯿﯿﻦ ﻫﻮﯾﺖ ﺷﺪﻧﺪ. اﻟﮕﻮی ﻣﻘﺎوﻣﺖ آﻧﺘﯽ ﻣﯿﮑﺮوﺑﯽ ﺑﻪ روش انتشار دیسک (ﮐﺮﺑـﯽ- بائر) اﻧﺠـﺎم ﮔﺮﻓـﺖ. درصد مقاومت ایزولههای کلبسیلاپنومونیه نسبت به سفکسیم 82%̨ سفوتاکسیم 81%̨ سفتریاکسون 73%̨ سفتازیدیم 72%̨ آمیکاسین 63%̨ آزیترومایسین60%̨ تتراسیکلین و نالیدیکسیک اسید 59%̨ جنتامسین 58% و ایمیپنم 43% تعیین گردید. نتایج به دست آمده نشان داد که ایمیپنم با کمترین درصد مقاومت در مقابل تمام ایزولههای کلبسیلا پنومونیه، موثرترین آنتیبیوتیک بوده است.

تعیین گروههای فیلوژنتیک اشریشیاکلیهای جدا شده از عفونت کلی باسیلوزیس طیور با استفاده از روش Multiplex-PCR
در میان بیماریهای ایجاد شده توسط اشریشیا کلی، نوعی بیماری سیستمیک حاد به نام کلیسپتیسمی وجود دارد که با حضور اشریشیا در خون، کلونیزه شدن در ارگانها شامل قلب، کبد و طحال مشخص میشود. هدف از این مطالعه طبقه بندی اشریشیا کلی جدا شده از عفونت کلیباسیلوزیس طیور و براساس گروهای فیلوژنتیک A ̨B1̨ B2 وD میباشد. در این مطالعه 80 سواپ اخذ شده از کبد و محوطه بطنی طیور بر روی محیط مک کانکی آگار کشت داده شدند. کلونیهای صورتی رنگ جداسازی شده و با انجام آزمونهای بیوشیمیایی به عنوان اشریشیا کلی تایید شدند. سپس با انجام Multiplex-PCR گروههای مختلف فیلوژنتیکی شناسایی گردیدند. از بین 80 نمونه 21 جدایه به عنوان اشریشیا کلی شناسایی شدند. 8 جدایه متعلق به گروه A (38 %)، 2 جدایه متعلق به گروه B1 (5/9 %)، 6 جدایه متعلق به گروه B2 (6/28 %) و 5 جدایه متعلق به گروه D2 (8/23 %) بودند. بر اساس نتایج مطالعه حاضر گروههای مختلف فیلوژنتیک در گلههای مرغ مادر مشاهده گردید. اغلب آنها در گروه A قرار گرفتند که به عنوان همزیست مطرح میباشند. مطالعات نشان میدهد که اشریشیا کلی پاتوژنیک دارای میزان مقاومت آنتیبیوتیکی قابل ملاحظهای میباشد که ممکن است به طرق مختلف به گلههای گوشتی انتقال یابد و منجر به تحمیل خسارت اقتصادی به صنعت طیور میگرد. بنابراین، گامهای مهمی باید جهت ریشهکنی اشریشیا کلی پاتوژنیک برداشته شود.

تعیین گروه های فیلوژنیکی باکتریهای اشریشیا کلی جدا شده از طیور گوشتی مبتلا به کلی باسیلوز در شهرستان زابل
باکتری اشریشیا کلی یک ارگانیسم فرصت طلب است که میتواند در طیور منجر به سندرمهای کلیباسیلوز (کلیسپتیسمی، پریکاردیت، پری هپاتیت و سالپنژیت و...) گردد. اشریشیا کلی دارای 4 گروه فیلوژنیکی شامل A، B1، B2 و D است. فیلوتایپینگ ایزولههای اشریشیا کلی روشی مناسب جهت بررسی نوع پراکندگی گروههای فیلوژنیک ایزولههای اشریشیا کلی در مناطق مختلف میباشد. بررسی حاضر به منظور تعیین گروههای فیلوژنیک اشریشیا کلی در شهرستان زابل صورت گرفت، در این مطالعه به منظور تعیین فراوانی گروههای فیلوژنیکی باکتری اشریشیاکلی جدا شده از طیور گوشتی زابل، 144 قطعه جوجه گوشتی مشکوک به کلی باسیلوز نمونه گیری شد و در محیط TSBبه آزمایشگاه منتقل گردید. با انجام کشتهای میکروبی و آزمایشهای بیوشیمایی رایج تعداد 100 ایزوله اشریشیا کلی تعیین هویت گردید، DNA باکتریهای ایزوله شده به روش جوشاندن استخراج گردید و به روش triplex PCR گروههای فیلوژنیک آنها تعیین شد. در این مطالعه از مجموع 100 ایزوله، 36 % ، 27 %، 23 % و 14 % به ترتیب در گروههای فیلوژنیک B1، D، A و B2 قرار گرفتند. با انجام مطالعه حاضر مشخص گردید که اغلب ایزولههای اشریشیا کلی جدا شده از طیور گوشتی مبتلا به کلیباسیلوز در شهرستان زابل متعلق به گروههای فیلوژنیک B1 بودند.

تفاوت دوگروه فیلوژنتیکی A و B2 سویههای اشریشیا کلی یوروپاتوژنیک از نظر توزیع ژنهای حدت
اشریشیا کلی به عنوان فراوانترین باکتری ایجاد کننده عفونت ادراری معرفی شده است. سویههای اشریشیا کلی ایجاد کننده عفونت ادراری که به عنوان «یوروپاتوژنتیک» شناخته میشوند، حاوی فاکتورهای حدت متنوع میباشند. بر اساس مطالعات قبلی سویههای متعلق به گروه فیلوژنتیکی B2 به عنوان مهمترین سویه، درحالیکه سویههای گروه Aبه عنوان کم اثرترین سویه در ایجاد عفونت ادراری مطرح هستند. در این مطالعه تعداد 100 نمونه اشریشیا کلی جدا شده از بیماران مبتلا به عفونت ادراری با روشهای استاندارد بیوشیمیایی تایید شد. پس از استخراج DNA ژنومی با روش Triplex-PCR تعداد 72 سویه ( 55 سویه متعلق به گروه فیلوژنتیکی B2 و 17 نمونه متعلق به گروه A) برای تعیین میزان توزیع ژنهای حدت انتخاب شدند. فراوانی ژنهای cnf1،irp2،ihaوompT به ترتیب به میزان 39%، 29% ، 92% و 78% مشاهده شد. میزان فراوانی این ژنها در گروه فیلوژنتیکی B2 به مراتب از گروه A بیشتر بود. تفاوت معنیداری در میزان توزیع ژنهای cnf1 و irp2 در دو گروه فیلوژنتیکی B2 و A مشاهده شد ( 0.05 P≤). از نظر الگوی توزیع ژنی 10 الگوی منحصر به فرد (Ec1-Ec10 ) برای این دو گروه مشاهده شد. نتایج این مطالعه نشان داد که سویههای B2 حاوی ژنهای حدت بیشتری نسبت به سویههای A هستند و احتمالا نقش مهمتری در ایجاد عفونت ادراری دارند.

شناسایی و تایپینگ استافیلوکوکوس اورئوس جدا شده از موارد ورم پستان بالینی گاوی در شهرستان سنندج بر اساس آنالیز PCR-RFLPژن aroA
استافیلوکوکوس اورئوس یکی از مهمترین عوامل ایجادکننده ورم پستان عفونی در دامهای اهلی است. به دلیل وجود سویههای متعدد استافیلوکوکوس اورئوس و تفاوتهای سویهای در بازآرایی آللهای کروموزمی، روشهای ژنوتایپینگ متفاوتی همچون آنالیز DNA کروموزمی توسط هضم آنزیمی به منظور تایپینگ ژنتیکی باکتری معرفی شدهاند. در بررسی حاضر، برای اولین بار تنوع ژنتیکی سویههای استافیلوکوکوس اورئوس جداشده از موارد ورم پستان بالینی گاوی در شهرستان سنندج براساس آنالیز PCR-RFLP ژن aroA ارزیابی گردید. در این تحقیق تعداد 120 نمونه شیر ورم پستان گاوی به روش استریل جمعآوری و بررسی گردید. استافیلوکوکوس اورئوس جداشده به روش کشت و آزمایشات روتین باکتریولوژیکی، با روش PCR مبتنی بر ژن aroA آنالیز شد. قطعه حاصل از تکثیر با اندازه 1153 جفت باز توسط آنزیم تعیین حدودی TaqI هضم و قطعات ایجاد شده الکتروفورز شدند. 28 سویه استافیلوکوکوس اورئوس در روش فنوتیپی جداسازی و قطعه موردنظر در تمام جدایهها مشاهده شد. در هضم آنزیمی، دو نوع الگوی هضم قابل نامگذاری براساس مطالعات قبلی ایجاد شد. ژنوتیپ B در 23 مورد (82٪) و ژنوتیپ N در پنج مورد (۱8٪) شناسایی شد. نتایج نشان میدهند که استافیلوکوکوس اورئوس در ایجاد ورم پستان بالینی در سنندج دخیل بوده و علیرغم وجود ژنوتیپهای محدود، تنوع سویهای در جدایههای این باکتری در منطقه وجود دارد. عدم شناسایی ژنوتیپ جهانی A و نیز ژنوتیپ اختصاصی باکتری در سنندج، برای اولین بار گزارش میشود.

مطالعه اثرات ضدمیکروبی عصاره اتانولی گیاه علف مار بومی سیستان بر روی سالمونلا تیفیموریوم مقاوم به آنتیبیوتیک
امروزه خواص ضد باکتریایی عصاره گیاهان دارویی به عنوان یک جایگزین مناسب برای آنتیبیوتیکها مورد توجه است. هدف از این مطالعه بررسی اثرات ضدمیکروبی عصاره برگ گیاه علف مار بر روی سالمونلا تیفیموریوم (Salmonella typhmurium) جدا شده از طیور در شرایط آزمایشگاهی بود. عصاره اتانولی گیاه علف مار با استفاده از دستگاه خلاء از مرکز (روتاری) استخراج شد. همچنین تعداد 12 سویه باکتری سالمونلا تیفیموریوم نیز از طیور زابل جداسازی شد. حداقل غلظت بازدارندگی و حداقل غلظت کشندگی عصاره الکلی گیاه در غلظتهای مختلف با روش رقتسازی در چاهک بر روی باکتریها تعیین شد. حساسیت سویهها به آنتیبیوتیکهای مختلف نیز با روش استاندارد دیسک دیفیوژن کربی بائر مورد ارزیابی قرار گرفت. نتایج حاصل از این مطالعه نشان داد که سویههای سالمونلا تیفیموریوم به آنتی بیوتیکهای پنیسیلین (% 100)، آمپیسیلین (% 100)، تتراسیکلین (% 6/16) و آمیکاسین (% 3/8) مقاوم میباشد. همچنین، عصاره اتانولی گیاه علف مار اثر مهارکنندگی قابل توجهی در برابر سالمونلا تیفیموریوم از خود نشان داده است. نتایج حاصل از این مطالعه نشان داد که امکان استفاده از عصاره اتانولی گیاه علف مار به عنوان جایگزین مناسب آنتیبیوتیکهای متداول وجود دارد.
