مقایسه‌ی الگوی مقاومت آنتی‌بیوتیکی در ایزوله‌های سالمونلا تیفی‌موریوم و اشریشیاکلی جداشده از محتویات روده و پوست نزدیک به پرینه در سگ‌های استان اصفهان

چکیده

سالمونلا و اشریشیاکلی از عوامل مهم عفونت‌های روده‌ای و اسهال می‌باشند. افزایش مقاومت سالمونلا و اشریشیاکلی نسبت به آنتی‌بیوتیک‌های رایج به‌عنوان یک مشکل جهانی مطرح است. از آنجایی‌که سگ‌های خانگی به‌عنوان منابع بالقوه‌ی این دو باکتری مطرح می‌باشند، مطالعه‌ی حاضر با هدف بررسی الگوی مقاومت آنتی‌بیوتیکی باکتری‌های سالمونلا تیفی‌موریوم و اشریشیاکلی جداشده از محتویات روده و پوست نزدیک به پرینه در سگ‌های استان اصفهان انجام شد. سواب‌های رکتال و پوستی اخذ شده از 100 سگ ارجاعی به کلینیک‌های حیوانات خانگی در شهر اصفهان کشت داده شدند. پس از کشت میکروبی و تائید مولکولی جدایه‌های سالمونلا تیفی‌موریوم و اشریشیاکلی، حضور شایع‌ترین ژن‌های کدکننده مقاومت آنتی‌بیوتیکی در این جدایه‌ها به روش PCR و الگوی فنوتیپی مقاومت آنتی‌بیوتیکی به روش انتشار دیسک ارزیابی شدند. تمام جدایه‌های اشریشیاکلی جداشده از محتویات روده به جنتامایسین، تتراسایکلین، سیپروفلوکساسین و تری متوپریم-سولفامتوکسازول مقاوم بودند. همچنین همه‌ جدایه‌های سالمونلا تیفی‌موریوم جداشده از محتویات روده نسبت به تتراسایکلین مقاومت نشان دادند. در تمام جدایه‌های اشریشیاکلی جداشده از محتویات روده ژن dfrA1 ردیابی شد. شایع‌ترین ژن ردیابی شده در جدایه‌های سالمونلا تیفی‌موریوم جداشده از محتویات روده tetB  و dfrA1، (80 درصد) بودند. مقاومت آنتی‌بیوتیکی چندگانه در این جدایه‌ها نشان‌دهنده استفاده بی‌رویه از آنتی‌بیوتیک‌ها بوده و لزوم انجام آنتی‌بیوگرام جهت انتخاب مؤثرترین آنتی‌بیوتیک در درمان بیماری‌ها را بیش ‌از پیش نشان می‌دهد.

چکیده [English]

Salmonella and Escherichia coli are important bacteria that cause intestinal infections and diarrhea. The increment of salmonella and E.coli antibiotic resistance is considered as a major global problem. Since indoor dogs are known as potential reservoirs for these two bacteria, this study was designed for investigation of antibiotic resistance pattern of salmonella typhimurium and Escherichia coli isolated from rectal and perineal region skin swabs of these dogs in Esfahan province. Skin and rectal swabs obtained from 100 dogs referred to veterinary clinics, were cultured for bacterial isolation. After bacterial culture and isolation, salmonella and E.coli positive colonies were confirmed using Polymerase chain reaction. Then the presence of the most prevalent antibiotic resistance genes were investigated using PCR method and phenotypic patterns were determined using disc diffusion method. All of the E.coli bacteria isolated from rectal samples were resistant to gentamycin, tetracycline, ciprofloxacin, and trimethoprim- sulfamethoxazole. Furthermore, all of salmonella rectal isolates were resistant to tetracycline. dfrA1 gene was detected in all of E.coli bacteria isolated from rectal swabs. Tet B and dfrA1 were the most detected genes in salmonella typhimurium bacteria isolated from rectal swabs. Multiple antibiotic resistance in these isolates shows extra usage of antibiotics and to overcome this problem, antibiogram test is suggested to choose the best antibiotic for the treatment of infectious disease.
