بررسی اثرات ضد میکروبی عصاره‌های اتانولی، متانولی و اتیل استات کلپوره (Teocrum polium) بر روی استافیلوکوکوس اورئوس جدا شده از گوسفند

چکیده

باکتری گرم مثبت استافیلوکوکوس اورئوس(Staphylococcus aureus) از جمله رایج‌ترین عوامل شیوع مسمومیت‌های غذایی باکتریایی است و بین انسان و دام شایع است. بر همین اساس، هدف مطالعه حاضر جداسازی باکتری استافیلوکوکوس اورئوس از بینی گوسفندان بومی منطقه سیستان و بررسی مقاومت آنتی‌بیوتیک سویه‌های جداسازی شده به آنتی‌بیوتیک‌های مرسوم بود. همچنین اثر ضد میکروبی عصاره‌های اتانولی، متانولی و اتیل استات گیاه کلپوره (Teocrum polium) بر روی سویه‌های استافیلوکوکوس اورئوس بررسی شد. برای این منظور گیاه کلپوره از منطقه سیستان جمع‌آوری گردید. تهیه عصاره به کمک دستگاه روتاری و با کمک روش تقطیر در خلاء انجام گرفت. تعداد 10 سویه باکتری استافیلوکوکوس اورئوس از بینی گوسفندان بومی منطقه سیستان به کمک روش‌های بیوشیمیایی جداسازی شد. با استفاده از روش استاندارد دیسک دیفیوژِن کربی- بائر، حساسیت سویه‌های باکتری نسبت به آنتی‌بیوتیک‌های اکساسیلین، آموکسی سیلین-کلاولانیک اسید، آمپی‌سیلین، جنتامایسین، سفازولین و ونکومایسین مورد ارزیابی قرار گرفت. حداقل غلظت مهارکنندگی و حداقل غلظت کشندگی عصاره‌ها در تیمار با عصاره‌های گیاه کلپوره با استفاده از روش رقت‌سازی در چاهک تعیین شد. نتایج مطالعه حاضر نشان داد که هر سه عصاره اتانولی، متانولی و اتیل استات درجات متفاوتی از مهارکنندگی را بر روی 10 سویه باکتری جداسازی شده دارند. با این وجود عصاره اتیل استات گیاه کلپوره در غلظت‌های پایین‌تری خاصیت مهارکننده بر روی بیشتر سویه‌های باکتری جداسازی شده داشت. همانند خاصیت مهارکنندگی قوی‌تر عصاره اتیل استات کلپوره، این عصاره در مقایسه با عصاره‌های اتانولی و متانولی خواص کشندگی قوی‌تری نیز علیه باکتری استافیلوکوکوس اورئوس نشان داد. نتایج مطالعه حاضر نشان داد عصاره اتیل استات گیاه کلپوره می‌تواند جایگزین مناسبی برای آنتی‌بیوتیک‌های متداول باشد.

چکیده [English]

Staphylococcus aureus is a gram positive Bactria which has been considered as one of the most common agent of food bacterial poisoning and also considered as a zoonotic disease. Therefore, the aim of the current study was to isolate of different strains of S. aureus from nasal samples of sheep from Sistan region and evaluation of antibiotic resistance of the isolated bacteria against common antibiotics. Moreover, the antibacterial activity of ethanol, methanol and ethyl acetate extracts of Teucrium polium plant on S. aureus was assessed. The plant samples were gathered from Sistan Region. Ethanol extract of plant samples was extracted using vacuum from the center (rotary) apparatus. A total number of 10 S. aureus strains were isolated from the sheep nasal samples from Sistan region. Susceptibility to Oxacillin, Amoxicillin-clavulanic acid, ampicillin, Gentamicin, Cefazolin and Vancomycin antibiotics was evaluated by the Kirby-Bauer disk diffusion standard method. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of different concentrations of plant extract were determined on bacteria using dilution in well method. Results of the current study explained that all of the three ethanol, methanol and ethyl acetate extracts had various bactericidal levels on 12 isolated bacteria. Nevertheless, the ethyl acetate extract was batter than two other extracts due to its lower concentrations for its MIC and MBC against S. aureus. Results derived from the current study suggest that ethyl acetate extract of T. polium would be a suitable alternative for common antibiotics.
