بررسی فعالیت آنتی‌باکتریایی فیلم‌های برپایه کربوکسی‌متیل سلولز ترکیب‌شده با نانوذرات اکسید روی و عصاره پوست انار برای نگهداری نمونه‌های ماهی قزل‌آلای رنگین‌کمان

چکیده
ماهی قزل‌آلا به فساد میکروبی بسیار حساس است و امروزه استفاده از بسته‌بندی‌های زیست‌فعال برای افزایش ماندگاری مواد غذایی فسادپذیر مورد توجه قرار گرفته است. هدف از این مطالعه بررسی تأثیر فیلم‌های زیست‌فعال بر پایه کربوکسی‌متیل سلولز (CMC)  ترکیب ‌شده با نانوذرات اکسید روی (ZnO) و عصاره پوست انار (PE) بر حفظ کیفیت ماهی قزل‌آلای رنگین‌کمان بود. فیلم‌ها به روش ریخته‌گری محلول تهیه شدند و در پنج گروه مختلف ارزیابی شدند. فیلم‌ها شامل کربوکسی‌متیل سلولز (CMC)، کربوکسی‌متیل سلولز/ نانوذرات اکسید روی (CMC/ZnO)، کربوکسی‌متیل سلولز/ نانوذرات اکسید روی/50/0درصد عصاره پوست انار (CMC/ZnO/0.5% PE)، کربوکسی‌متیل سلولز/ نانوذرات اکسید روی/ 1درصد عصاره پوست انار (CMC/ZnO/1% PE)  و کربوکسی‌متیل سلولز/ نانوذرات اکسید روی/ 2درصد عصاره پوست انار (CMC/ZnO/2% PE)  بودند. نمونه‌های ماهی در دمای 25 درجه‌ی سانتی‌گراد برای مدت یک هفته نگهداری شدند. ویژگی‌های کیفی نمونه‌های ماهی قزل‌آلا شامل میزان بازهای ازته فرار (TVBN)، میزان کل باکتری‌های زنده (TVC) و pH بررسی شد. نتایج نشان داد که فیلم‌های حاوی نانوذرات اکسید روی و عصاره پوست انار به‌طور معنی‌داری (P<0.05) موجب کاهش TVBN و TVC شدند (P<0.05). همچنین،pH  نمونه‌ها در گروه‌های تیمار شده با فیلم‌ها نسبت به گروه کنترل به‌طور قابل توجهی پایین‌تر بود (P<0.05) که نشان‌دهنده‌ی کارایی این فیلم‌ها در کاهش فعالیت‌های میکروبی و تأخیر در روند فساد ماهی قزل‌آلای رنگین‌کمان بود. این نتایج نشان می‌دهند که استفاده از فیلم‌های کربوکسی‌متیل سلولز ترکیب‌شده با نانوذرات اکسید روی و عصاره پوست انار، به‌ویژه فیلم CMC/ZnO/2% PE، می‌تواند به‌عنوان روشی مؤثر برای حفظ کیفیت و افزایش ماندگاری ماهی قزل‌آلا در طول نگهداری باشد.

Antibacterial activity of carboxymethyl cellulose-based films combined with zinc oxide nanoparticles and pomegranate peel extract for preservation of Rainbow trout samples

چکیده English

Rainbow trout is highly susceptible to microbial spoilage, and use of active packaging to extend the shelf life of perishable foods has gained considerable attention in recent years. The aim of this study was to investigate the effect of bioactive films based on carboxymethyl cellulose (CMC) combined with zinc oxide nanoparticles (ZnO) and pomegranate peel extract (PE) on maintaining the quality of rainbow trout. Films were prepared by the solution casting method and evaluated in five different groups. The films included carboxymethyl cellulose (CMC), carboxymethyl cellulose/zinc oxide nanoparticles (CMC/ZnO), carboxymethyl cellulose/zinc oxide nanoparticles/0.5% pomegranate peel extract (CMC/ZnO/0.5% PE), carboxymethyl cellulose/zinc oxide nanoparticles/1% pomegranate peel extract (CMC/ZnO/1% PE), and carboxymethyl cellulose/zinc oxide nanoparticles/2% pomegranate peel extract (CMC/ZnO/2% PE). The fish samples were stored at 25°C for one week. The quality characteristics of the rainbow trout samples, including the total volatile basic nitrogen (TVBN) content, total viable count (TVC), and pH, were evaluated. The results showed that films containing zinc oxide nanoparticles and pomegranate peel extract significantly reduced TVBN and TVC (P<0.05). Additionally, the pH of the samples in the treated film groups was significantly lower than that in the control group (P<0.05), indicating the efficacy of these films in reducing microbial activity and delaying the spoilage of rainbow trout. These results suggest that the use of carboxymethyl cellulose-based films combined with zinc oxide nanoparticles and pomegranate peel extract, especially the CMC/ZnO/2% PE film, could be an effective method for maintaining quality and extending the shelf life of rainbow trout during storage.
