بررسی اثر اسانس اژنول روی تک‌یاخته تریکوموناس گالینه در شرایط آزمایشگاهی و تجربی

چکیده
در این مطالعه اثرات اسانس اژنول که ماده‌ی مؤثر گیاه میخک می‌باشد روی انگل تریکوموناس گالینه در شرایط آزمایشگاهی و تجربی مورد بررسی قرار گرفت. برای انجام مطالعه آزمایشگاهی از محیط کشت تغییر یافته دیاموند استفاده شد. نمونه انگل از چینه‌دان کبوتر آلوده به این انگل تهیه شد. پس از کشت انگل به دو گروه: گروه یک بدون تیمار یا گروه کنترل و گروه دو، گروه تیمار شده با اسانس اژنول تقسیم شد و برای انجام این مطالعه از پلیت 96 خانه استفاده شد. محیط کشت حاوی انگل و اسانس اژنول به این پلیت‌ها اضافه شد و در نهایت غلطت 10، 5، 5/2، 25/1 میکروگرم در میلی‌لیتر اسانس اژنول به دست آمد و در زمان‌های 6، 12، 24 و 48 ساعت پس از تیمار با اژنول تعداد تک‌یاخته تریکوموناس گالینه زنده و مرده محاسبه شد. در مطالعات تجربی تعداد 60 کبوتر به گروه‌های تیمار با اسانس اژنول 10 میلی‌گرم در هر کیلوگرم و گروه کنترل تقسیم شد و سپس هر روز تا 5 روز روزی یک‌بار به‌صورت خوراکی تحت درمان قرار گرفتند. نتایج نشان می‌دهد در مطالعات آزمایشگاهی اسانس اژنول در دوز 10 میکروگرم بر میلی‌لیتر بعد از گذشت 48 ساعت سبب مرگ کامل انگل تریکوموناس گالینه می‌شود. در مطالعات تجربی نشان می‌دهد 5 روز پس از درمان با اسانس اژنول 10 میلی‌گرم در هر کیلوگرم وزن بدن هیچ انگل تریکوموناس گالینه زنده‌ای در این کبوتر مشاهده نمی‌شود. در نتیجه این مطالعات نشان می‌دهد که اسانس اژنول می‌تواند روی انگل تریکوموناس گالینه مؤثر باشد.

This study sought to explore the in vitro and in vivo efficacy of eugenol, the principal compound in the clove plant, against Trichomonas gallinae (T. gallinae). In the in vitro investigation, Diamond's modified culture medium was employed. The parasite sample was derived from an infected pigeon, cultured, and then maintained in an incubator at 37 °C for 48 hours until the protozoa count reached approximately 1×104 parasites per ml of culture medium. Subsequently, two groups were assigned: Group one received no treatment, while group two was treated with eugenol. Ninety-six plates were utilized, and the culture medium containing the parasite and eugenol was added to these plates. Doses of 10, 5, 2.5, and 1.25 µg/ml of eugenol were administered. The number of dead and alive parasites, along with the percentage of parasite mortality, was measured at intervals of 6, 12, 24, and 48 hours post-mixing. For the in vivo experiment, 60 pigeons were experimentally infected with T. gallinae and divided into two groups. Group one served as the control, receiving no treatment, while group two was treated with eugenol at a dose of 10 mg/kg for five consecutive days. Results from the in vitro study demonstrated that a concentration of 10 μg/ml of eugenol resulted in 100% lethality in T. gallinae after 48 hours. The in vivo study indicated that eugenol at a dose of 10 mg/kg led to 100% recovery five days post-treatment. These findings underscore the effectiveness of eugenol against the T. gallinae parasite, suggesting its potential as an alternative treatment.
