اپیدمیولوژی مولکولی، تعیین ژنوتیپ و مطالعه فیلوژنتیکی نوروویروس گاوی در گوساله‌های مبتلا به اسهال در ایران

چکیده
نورویروس، به‌ویژه نورویروس‌های گاوی (BoNoV)، ویروس‌های RNA دار و عامل گاستروانتریت در انسان و دام هستند و برخی ژنوتیپ های آن در اسهال گوساله‌ها نقش دارند. بیماری‌های گوارشی در گوساله‌های جوان می‌تواند خسارات اقتصادی قابل توجهی ایجاد کرده و بهره‌وری گله‌های گاو شیری را تحت تأثیر قرار دهد. نورویروس ها قادرند گونه‌های مختلفی از جمله گاو، خوک، سگ، گربه، موش، و انسان را آلوده کنند. نورویروس‌های حیوانی (مانند نورویروس گاوی، خوکی) از نورویروس انسانی متمایز هستند و شواهد قطعی در مورد عفونت متقاطع بین گونه‌ها کم است . همچنین، اطلاعات علمی درباره احتمال آلودگی شیر با نورویروس بسیار اندک گزارش شده است. در این مطالعه، شیوع و ویژگی‌های ژنتیکی نورویروس گاوی به‌عنوان یکی از عوامل گاستروانتریت در گوساله‌های ایران بررسی شد. در مجموع، ۱۶۳ نمونه مدفوع از گوساله‌های اسهالی کمتر از دو ماه از پنج استان کشور طی سال‌های ۲۰۱۹ تا ۲۰۲۰ جمع‌آوری و با روش RT-PCR و استفاده از دو جفت پرایمر تحلیل شدند. با توجه به محدودیت منابع، تنها دو نمونه برای انجام آنالیزهای فیلوژنتیک مورد توالی‌یابی قرار گرفتند. داده‌های توالی در حال حاضر با شماره‌های SUB13053245 و SUB13053259 در GenBank ثبت شده‌اند. نتایج RT-PCR نشان داد که ۶۸ نمونه (۴۱٪) برای ژنوتیپ III نورویروس گاوی مثبت بودند. همچنین، فراوانی موارد مثبت در فصول سرد سال (پاییز و زمستان) بیشتر بود. تحلیل فیلوژنتیک نشان داد که نمونه SUB13053245 شباهت ژنتیکی بالایی (۹۷٪) با سویه‌های گزارش‌شده از کشورهای شرق آسیا و مصر دارد؛ در حالی‌که نمونه SUB13053259 بیشترین نزدیکی ژنتیکی را با سویه‌های کشورهای اروپایی مانند بلژیک، فرانسه و انگلستان نشان داد. این یافته‌ها بیانگر شیوع بالای ژنوتیپ III نورویروس گاوی در گوساله‌های اسهالی در ایران است. همچنین، نتایج بر ضرورت انجام مطالعات گسترده‌تر در چارچوب رویکرد «یک‌بهداشت» (One Health) برای درک بهتر اپیدمیولوژی نورویروس‌های حیوانی و بررسی احتمال انتقال بین‌گونه‌ای تأکید دارد.

Molecular epidemiology, genotyping and phylogenetic study of bovine norovirus from diarrheic calves in Iran

Norovirus is a highly contagious foodborne pathogen associated with calf diarrhea, causing economic losses in dairy cattle herds. This virus can infect a wide range of species, including cattle, pigs, dogs, cats, mice, sheep, lions, and humans. In this study, the prevalence and genetic sequencing of bovine norovirus as a causative agent of gastroenteritis in calves in Iran were investigated. A total of 163 fecal samples were collected from diarrheic calves under two months of age across five provinces in Iran during 2023–2024 and analyzed using RT-PCR with two pairs of primers. Two positive samples were sequenced for phylogenetic analysis (GenBank accession numbers: SUB13053245 and SUB13053259). RT-PCR results indicated that 68 samples (41%) tested positive for bovine norovirus genogroup III. Additionally, a higher prevalence of norovirus was observed in colder seasons (autumn and winter). Phylogenetic analysis revealed that sample SUB13053245 exhibited a close genetic relationship (97% similarity) with bovine norovirus strains reported from East Asia and Egypt, while sample SUB13053259 clustered with norovirus strains from European countries such as Belgium, France, and the United Kingdom. These findings highlight the high prevalence of bovine norovirus genogroup III in diarrheic calves in Iran, emphasizing the need for further investigations, particularly within the One Health framework, to assess the molecular epidemiology and risk factors associated with norovirus infections in Iran.
